Job DescriptionThe Maduke laboratory at Stanford University seeks a postdoctoral fellow to work on a collaborative interdisciplinary project to develop small-molecule chloride-channel inhibitors as potential new therapeutics for hyponatremia.
The CLC (“Chloride Channel”) family of transporters and channels are ubiquitous in all organisms. Among them, CLC-Ka and CLC-Kb are essential for Cl– and water handling in the kidney. CLC-Ka is localized to the thin ascending limb, where it helps to establish a steep solute gradient that drives renal water reabsorption. As such, CLC-Ka is a potential drug target for treating pathologic water retention (hyponatremia) that frequently complicates the management of patients with hypertension, heart failure, or cirrhosis. Hyponatremia affects up to 75% of patients with serious illness and 1.7% of the general population. In hyponatremia, the excess water causes potentially dangerous swelling, particularly in the brain. Symptoms include muscle weakness and cognitive impairment, and severe consequences can include irreversible brain damage or even death. Currently, the only type of medication that exists for treating hyponatremia contains substantial drawbacks due to potential for causing life-threatening liver damage. In this project, we seek to develop molecules that can modulate the function of CLC-Ka, with the long-term goal of translating this research to create novel therapeutic treatments for hyponatremia.
The project will involve structure-activity relationship (SAR) studies to optimize potency, selectivity, and pharmacokinetic properties of small-molecule CLC-Ka inhibitors. A Ph.D. in chemistry, biophysics, physiology, or related field is required. Experience with electrophysiology and/or synthetic organic chemistry is desired. However, highly motivated candidates with a strong background in the physical sciences will also be considered. An ability to think quantitatively and to communicate clearly is essential.
